چکیده

BACKGROUND AND PURPOSE Bacteriophages, viruses that cause lysis of bacteria, can serve as an effective tool for the control of infections, especially those caused by multiple drug-resistant organisms. Klebsiella pneumoniae is one of the most predominant pathogens associated with burn wound infections, causing considerable morbidity and mortality. This study investigated the protective effect of K. pneumoniae-specific bacteriophage KØ1 isolated from the environment in a mouse model of burn wound infection induced by K. pneumoniae. METHODS The toxicity of the bacteriophage and its in vivo survival and stability in mice was assessed. The fatal dose of K. pneumoniae was ascertained in third-degree burn wound infection. The protective effect of the bacteriophage administered via the subcutaneous or intraperitoneal route was investigated. RESULTS A substantial decrease in the bacterial load of blood, peritoneal lavage, and lung tissue was noted following treatment with the bacteriophage preparation. The decrease in microbial count was evident when the bacteriophage was administered either via the subcutaneous or intraperitoneal route. CONCLUSION These results suggest that bacteriophages have the potential to modulate the course of burn wound infection caused by K. pneumoniae.

Protection against fatal Klebsiella pneumoniae burn wound sepsis by passive transfer of anticapsular polysaccharide.

Klebsiella pneumoniae KP1-0 capsular polysaccharide (PS) was isolated from culture supernatants by coprecipitation with N-cetyl-N,N,N-trimethylammonium bromide. PS was composed primarily of carbohydrate and contained less than 1% (wt/wt) protein and nucleic acids. The protective capacity of passively transferred anti-PS was evaluated in a murine burn wound sepsis model. Anti-PS antibody was fou...

Topical treatment of Klebsiella pneumoniae B5055 induced burn wound infection in mice using natural products.

BACKGROUND Burn wound infection remains the principal cause of death in burn patients. Efficacy of honey and aloe vera gel was evaluated in the treatment of burn wound infection induced with Klebsiella pneumoniae B5055 and their efficacy was compared with an isolated and well-characterized Klebsiella specific phage Kpn5. METHODOLOGY A full thickness burn wound was induced in mice and infected...

Efficacy of bacteriophage treatment in murine burn wound infection induced by klebsiella pneumoniae.

In the present study, the therapeutic potential of purified and well-characterized bacteriophages was evaluated in thermally injured mice infected with Klebsiella pneumoniae B5055. The efficacy of five Klebsiella phages (Kpn5, Kpn12, Kpn13, Kpn17, and Kpn22) was evaluated on the basis of survival rate, decrease in bacterial counts in different organs of phage-treated animals, and regeneration o...

Experimental Klebsiella pneumoniae burn wound sepsis: role of capsular polysaccharide.

The role of Klebsiella pneumoniae capsular polysaccharide in relation to virulence in a murine burn wound sepsis model was investigated. Burn trauma markedly predisposed mice to lethal K. pneumoniae sepsis. A highly encapsulated variant (KP1-O) derived from K. pneumoniae KP1 was found to be extremely virulent for burned mice (50% lethal dose less than 10 organisms), whereas another variant (KP1...

Klebsiella pneumoniae alleviates influenza-induced acute lung injury via limiting NK cell expansion.

A protective effect induced by bacterial preinfection upon a subsequent lethal influenza virus infection has been observed, but the underlying immune mechanisms have not yet been fully elucidated. In this study, we used a mouse model of Klebsiella pneumoniae preinfection to gain insight into how bacterial preinfection influences the subsequent lethal influenza virus infection. We found that K. ...
